Our convention for storing and labeling your work is as follows:

Everyone's prefix is composed of their initials.  For example, Ramez Danial's prefix is rd (lowercase).

Minipreps = [PREFIX]M[#].  For example, rdM1, rdM2, rdM3

Gel Preps = [PREFIX]G[#].  For example, rdG1, rdG2, rdG3

Restriction Digests = [PREFIX]R[#].  For example, rdR1, rd2, rdR3

Frozen Bacteria = [PREFIX]F[#].  For example, rdF1, rdF2, rdF3.

Frozen Yeast = [PREFIX]Y[#].  For example, rdY1, rdY2, rdY3.

I suggest maintaining multiple backups of your work to insure against catastrophies:

1) Store a frozen stock in the -80C. This can be done by adding 345 uL of 80% glycerol + 1.5 mL of healthy cell stock (preferably in late log phase but early stationary phase or overnight is usually OK).

2) Store a miniprepped version of the plasmid from the frozen stock in the -20C.
