Subgroup Specific


Receptor Group: 

  • Look into making a plasmid table organizing all the plasmids that we will be using

miRNA Group: 

  • Look into contacting and setting up a meeting with Jake Beal
  • 12 miRNA may be a little too much - look into constructing sensors for individual up-regulated or down-regulated miRNA and then look into combining two or more of these sensors 
  • Contact Jeremy Gam about his miRNA sensing circuit 
    • start looking into getting the miRNA in order to start testing
  • What is the test bed? 
    • Cell that doesn't have any of the miRNA and add the siRNA for the up-regulated miRNA 
    • Mimicking the down-regulated "phenotype" will be harder 
  • Alzheimer's disease model - look into that 
  • Journal Club on the miRNA profile paper

Treatment Group: 

  • What we are currently doing: 
    • Looking into directly assaying BACE2 activity - kit available 
      • look into getting a smaller kit 
    • Fusing together BACE2 with YFP
    • Looking into miRNA that inhibit BACE1 expression
    • Practice transfection with a constitutive promoter and an inducible 
  • BACE1 has a Bsa1 site in the middle of it - look into how that would affect cloning strategy 
  • Look into potential cross-talk resulting from the miRNA that we are considering using to inhibit BACE1 production 
  • BLAST: pairwise alignment tool 

Antibody Group: 

  • "that is a lot of experiments; you should be afraid" 
  • gBlocks will be ordered today 
  • Document how the leader sequence was selected on the wiki

Shinjini: 

  • Specific cytokines that will help microglia go down the phagocytic pathway 
  • Cytokines are involved in many other processes - might not be realistic to target them for treatment 
  • Look into using using microglia as vector hosts for our treatment
