Stored in 1% glycerol in small centrifuge tubes in -80 degree freezer in between Chisholm and DeLong labs on the bottom shelf in white styrofoam box.
Put 200 ul into 20 ml of Tryptic Soy Broth (TSB) (See bleow for recipe)
Put on room temperature shaker (150 rpm) for 12-14 hours
Check optical density using spec in Polz lab:
-Turn on monitor
-Vis- turn on
-Check wavelengthset to 600 nm
-Add a blank (culture medium)
-Blank sample
-Add your sample
-Read sample
-OD should be 0.25-0.6
-When finished TURN OFF LAMP
-Turn of monitor
Add 2 ml of culture to 20 ml of Artificial Seawater (ASW) (See Recipe below)
-Leave in lab for 24 hours - happy motile bacteria should result
-Measure OD
