Phage display libraries are typically screened by affinity selection in ELISA plates. Currently (as of 5/23/2012) we carry capture plates for T7 from Novagen as well as polyclonal antibodies against pIII of M13 (or fd) from Novus Biologicals. The T7 capture plates are stored in the general -4 degree fridge and the antibodies are stored in the general -20 degree fridge.

I used the following protocol to create anti-fd plates. The plates I made caught about 55 % of phage when incubated with 100 uL of 1E9 TU/mL of f88/6mer library for 3 hours at 37 degrees. I suspect that tweeks could be made to this protocol to increase the capture capacity (for example, maybe not using blocking reagent). 

Protocol for preparing the anti-fd plates:

1. anti-pIII is at 3.8 mg/mL. Dilute this in TBS to make 10 ug/mL solution.

2. Put 100 uL of the solution into each of the wells of a 96 well plate. parafilm the plates (to prevent evaporation) and leave at 4 degrees overnight.

4.  wash plates 4 times with TBS.

5. fill with 200 uL of blocking solution (I used PBS with 1 % BSA).  parafilm the plates (to prevent evaporation) and leave at 4 degrees overnight.

6. wash plates 4 times with TBS.  Store dry at 4 degrees.
