To do today:
- Run DNA for gel extraction. Also look to see if other, verified DNA contaminated or not.
- Pick colonies fusions
- Transform
Tomorrow:
- Miniprep lots of fusions. Make cell stocks. Verify, get rid of extra stocks.
- pick colonies for pEXPR_hEF1a: LilrB2
Monday:
- Midiprep pEXPR_hEF1a: LilrB2 again. Make cell stock. THROW AWAY ALL OTHER STOCKS.
Is it worth doing the gel extraction? No.
Labeling is pretty bad. pEXPR_hEF1a:LilrB2: L23, L24, L33, L34 are verified. L2s and L3s are monoclonal. I don't know which one the Midiprep DNA came from. But given it came from one of these, no point in gel extracting. Confusion is bad.
DNA running:
HL, L23, L24, L33, L34, Midiprep.
Strangest contamination ever!
Transforming with L2 and L3 DNA (L24, L34).
The gel for L34 looks kind of sketch (little green dot underneath), but I ran it again, and it is fine.
