- Transform all LRs (hEF1a:PT, hEF1a:LT, hEF1a:TC, TREt:TC)
- Pick hEF1a:YC colonies
- Miniprep TREt:YC if it worked. Otherwise, wait for Brian. Worked.
- Test LilrB2TCSG4 digest in random places. Is the main LilrB2 stock missing a chunk? Preposterous. It's fine.
- PCR C term YFP? Nope, brian's looking into it.
- Midiprep inoculate TREt:YC
Digest LilrB2TCSG4 with NcoI and PstI seperately.
Digest TREt:YC with ApaI
TREt:YFP_Cofilin 1, 2, 4 verified. Will midiprep 2.
Looks like NcoI did not cut LilrB2_TCS. Probably the enzyme was causing problems, because it makes NO sense that the ends of LilrB2 are present but a chunk is missing from the middle. Also, the band from PstI is in the right length level. It must have been a gel problem. The lowest band is missing because it is actually 20bp long.
Ran PT (pENTR_PirBTCSG4) to make sure it isn't contaminated).
