Transformations

On 2014 June 28, transformations of new Golden Gates for CD79A+stop, CD79A-TCS-Gal4VP16, Syk+stop were done, as well as transformations for Gmab Heavy and Gmab Light.  No colonies were observed on any of the plates on 2014 June 29 at 9am.

Liquid culture

Started 4 liquid cultures each (5mL LB + 5uL kanamycin) for Lyn+stop, Lyn-mKate, Lyn-TEVp, Syk-eYFP, Syk-TEVp.  Blue colonies were observed on the plates in addition to white colonies, suggesting that the Golden Gate worked.  White colonies were picked and cultured.  Placed in shaking incubator (37C) at 3:30pm.  Removed from incubator to refrigerator at 8am.
