Creating a Glycerol Stock
- Fill cryotubes (orange screw caps) with 150 uL of 100% glycerol and autoclave en mass (Find an instructor for the autoclave)
- Restriction verify your construct and then take the 1 mL saved from liquid culture and add 850 uL of it to the cryotube
- Vortex to ensure mixing of the glycerol and cell culture
- Store in the -80
Pro Tips:
Avoid repeated use of the cryostock as viability decreases with every freeze/thaw cycle
Use the label maker for labeling and label well you probably won't touch these tubes for several months
